(1)藥效學:本品具有鎮痛、抗炎及解熱作用。本品通過抑制環氧酶使組織局部前列腺素的合成減少及抑制白細胞的趨化性和溶酶體酶的釋放而起到藥理作用。本品治療關節炎時的鎮痛、消腫等療效與吲哚美辛、阿司匹林、萘普生相似。但由于本品抑制環氧酶-2所需的濃度高于抑制環氧酶-1的濃度，因此胃腸道的不良反應較多。

(2)藥動學:口服吸收好。食物可降低吸收速度，但不影響吸收總量。血漿蛋白結合率高達90%以上。經肝臟代謝。半衰期平均為50小時(30~86小時)，腎功能不全患者半衰期延長。由于半衰期較長，一次給藥即可維持24小時的血藥濃度相對穩定，多次給藥易致蓄積。一次服藥20mg，3~5小時血藥濃度達峰值，血藥有效濃度為1.5~2μg/ml。血藥穩定濃度在開始治療后7~12天方能達到。66%自腎臟排泄，33%自糞便排泄，內有<5%為原形物。
